Butyrophilin-like 2 regulates site-specific adaptations of intestinal γδ intraepithelial lymphocytes

چکیده

Abstract Tissue-resident γδ intraepithelial lymphocytes (IELs) orchestrate innate and adaptive immune responses to maintain intestinal epithelial barrier integrity. Epithelia-specific butyrophilin-like (Btnl) molecules induce perinatal development of distinct Vγ TCR + IELs, however, the mechanisms that control IEL maintenance within discrete segments are unclear. Here, we show Btnl2 suppressed homeostatic proliferation IELs preferentially in ileum. High throughput transcriptomic characterization site-specific -KO reveals regulated antimicrobial response module ileal IELs. deficiency shapes specificities TCRγ/δ repertoire diversity During DSS-induced colitis, mice exhibit increased inflammation delayed mucosal repair colon. Collectively, these data suggest fine-tunes frequencies inflammatory cues. Hence, Btnl-mediated targeting may help dissect their immunological functions diseases with segment-specific manifestations.

Epithelia Use Butyrophilin-like Molecules to Shape Organ-Specific γδ T Cell Compartments

Many body surfaces harbor organ-specific γδ T cell compartments that contribute to tissue integrity. Thus, murine dendritic epidermal T cells (DETCs) uniquely expressing T cell receptor (TCR)-Vγ5 chains protect from cutaneous carcinogens. The DETC repertoire is shaped by Skint1, a butyrophilin-like (Btnl) gene expressed specifically by thymic epithelial cells and suprabasal keratinocytes. Howev...

Dynamic migration of γδ intraepithelial lymphocytes requires occludin.

γδ intraepithelial lymphocytes (IELs) are located beneath or between adjacent intestinal epithelial cells and are thought to contribute to homeostasis and disease pathogenesis. Using in vivo microscopy to image jejunal mucosa of GFP γδ T-cell transgenic mice, we discovered that γδ IELs migrate actively within the intraepithelial compartment and into the lamina propria. As a result, each γδ IEL ...

Cell Density Counts of the Intestinal Intraepithelial Lymphocytes in the Celiac Patients

Background: Increased number of intestinal intraepithelial lymphocytes (IELs) is a key histological finding in the diagnosis of celiac disease (CD); however, the number of IELs in celiac patients and healthy subjects may vary from one region to another. Additionally, there are some seronegative celiac patients with a borderline histology. Objective: To determin...

Butyrophilin-like 1 encodes an enterocyte protein that selectively regulates functional interactions with T lymphocytes.

Although local regulation of T-cell responses by epithelial cells is increasingly viewed as important, few molecules mediating such regulation have been identified. Skint1, a recently identified member of the Ig-supergene family expressed by thymic epithelial cells and keratinocytes, specifies the murine epidermal intraepithelial lymphocyte (IEL) repertoire. Investigating whether Skint1-related...

Intestinal intraepithelial lymphocytes: the plot thickens

O ver the last few years, the characterization of T lyraphocytes within the intestinal epithelium (ilEL) has revealed phenotypic and functional heterogeneity that rivals only that observed within the thymus. Of note in this context is that thymic and intestinal epithdia share the same embryologic origin in that they are both derived from endoderm. Several lines of evidence have led to the concl...
